Materials & Methods:
COS-7 cells were grown in Dulbecco’s modified Eagle’s medium (DMEM) enriched with 10% (v/v) FBS with 5% CO2 at 37 C on 35 mm petri dishes with No. 1 coverglass bottoms (MatTek Ashland MA). These cells are extremely flat and tightly adherent to the coverglass which makes them good candidates for TIRF microscopy.
