Immunofluorescence Microscopy—Cells (300000/dish) were cultured on 35-mm poly-D-lysine-coated glass-bottomed culture dishes (MatTek P35GC-1.5-14-C) for 24 h. Cells were fixed with 2% paraformaldehyde permeabilized with 0.2% Triton X-100 in phosphate-buffered saline for 10 min washed three times with 0.1% Tween 20 in phosphate-buffered saline followed
by 1% bovine serum albumin in phosphate-buffered saline for 30 min incubated with Pdpn antiserum (1:100) overnight at 4 °C washed and then labeled with goat anti-Syrian hamster IgG conjugated to Alexa Fluor 488 (Molecular Probes A21110).
