Abstract:
The reduced folate carrier (RFC) is a major folate transport system in mammalian cells. RFC is highly expressed in the intestine and believed to play a role in folate absorption.

Materials & Methods:
Confocal Imaging 198 Colocalization of hRFC with DYNLRB1 was performed using live cell confocal imaging 199 following transient transfection of the human intestinal epithelial HuTu-80 cells with 200 hRFC-GFP and DsRed-DYNLRB1 plasmids. Monolayers were grown on glass-bottomed 201 petri dishes (MatTek Inc Ashland MA) and imaging was performed using a Nikon C-1 202 confocal scanner head attached to Nikon inverted phase contrast microscope. 203 Fluorophores were excited by using a 488-nm line from an argon ion-laser and 543 nm 204 line from HeNe ion laser emitted fluorescence was monitored with a 515 30-nm band205 pass (GFP) or at 570 ± 50+-nm long-pass (DsRed) filter.
