Materials & Methods:
Cell culture and transfection N2a cells were maintained at 37 °C and 5% CO2 in D-MEM containing 4500 mg/l D-glucose and L-glutamine (Invitrogen) supplemented with 5% fetal calf serum (Perbio Science Deutschland Bonn Germany) 1% L-glutamine (PAA Laboratories GmbH Pasching Austria) 1% penicillin/streptomycin (PAA) 1% nonessential amino acids (aa) (PAA) and 1% sodium pyruvate (PAA). Transient transfections were carried out using the Effectene®4 Transfection Reagent Kit (Qiagen GmbH Hilden Germany). For localization studies 9000 cells and for dye uptake assays 15000 cells were seeded on glass bottom culture dishes (MatTek Corporation Ashland MA USA). Plasmids (300 ng) were transfected and cells studied after 48 h.
